Where are you testing the wire at? Try testing it right before the fuse, just after the fuse, just before the amp, and finally the amp terminal. This should narrow down where the problem is located.

yeah that sounds like a really good idea also try measuring resistance between the amp groung and a different ground(try a factory ground if possible) and make sure you cleaned the paint off the ground spot for your amp.

Dolike john said, track down the line. Before fuse, after fuse, distro block, or just downt he wire in various places. If it's 12v directly before the fuse and not after, then there is something wrong in that region.
