In most cases by tightening the screw you're adding preload to a spring that compresses a ball bearing to seal off boost pressure until the boost pressure is high enough to overcome the spring pressure. Loosening the screw reduces the boost needed to bypass the seal and flow to the wastegate actuator.

+1 for my MBC(joe p)in my opinion you just need to starting screwing it in and then out(sounds sexual--lol)and use your boost gauge to guide you..
