Depends on whether this is your initial registration, moving from another state, etc, etc. If you want to know for sure you need to call and talk to someone at a Hillsborough County tax collector's office.

if you don't have a tag to transfer expect at least $325.00

The last time I bought a tag for a vehicle i paid 160 with tax.... This was a few years back. You can end up spending about 300+ if you just bought a car (new or used). Tax, new tag, title in your name could = 300+

just the tag itself is almost 300 now not including taxes

I just paid 377 for new tags and registration. This was for an "even trade" so no tax was involved.

New tag is like 2fitty. Title fee is like 70 something, registration is like 45-55 for small car. Transferring tag is $7.
